2009年10月 Screening of Japanese Chuso samples for their anti-obesity and anti-oxidant activities

【Objective】
Cordyceps species are rare and valuable medicinal mushrooms.They have been used for prevention and treatment of various diseases,especially in Chinese traditional medicine for centuries. The aim of this study was to evaluate anti-oxidant and anti-obesity activities of Japanese Chuso species samples in vitro.

【Materials and Methods】
Fungi were collected in Tohoku district, Japan, and their spores were cultivated in culture medium. Freeze dried metabolic extracts of cultivated fungi spores were used in these experiments as samples. Anti-oxidant activity was determined by DPPH(2,2-diphenyl-1-picryl-hydrazyl) radical scavenging method and anti-obesity activity was estimated by BALB-DTNB method (lipase inhibition assay).

【Results and Discussion】
Among 30 samples tested, no sample showed strong anti-oxidant activity, but samples 23 (mixture) and 26 (mixture) and weak activities. Their IC50 values were 111.5 and 58.9 ug/ml, respectively. In anti-obesity test, Isaria japonica (IR=50.99%) and Cordyceps sp. Kobayashi (IR=49.75%) showed the strongest activities. These active samples may be expected to make contribution to prevention and treatment of ageing and lifestyle-related exogeneous obesity.

30種類の試料中、強い抗酸化作用を示す虫草はなかった。しかし試料混合エキス23と試料混合エキス26は弱いながら抗酸化作用が確認された。それぞれ有効濃度50%はIC50は111.5と58.9μg/mlであった。
抗肥満作用について、有効濃度比率IRはハナサナギタケでは50.99%、ミジンイモムシタケでは49.75%で、強い活性を示した。
これらの活性がある試料は老化予防、生活様式などによる外因的肥満に対して役立つ可能性が出てきた。
